【预期用途】
本产品仅供科研使用。本试剂盒采用双位点酶联免疫吸附分析法(ELISA)定量测定人类血清或血浆中的类胰岛素生长因子1(IGF-1)。为避免结合蛋白的干扰,本检测中需进行样本预处理。
【概述】
类胰岛素生长因子(IGF-I,IGF-II)是一类参与细胞生长代谢调节的多肽。在循环系统中,超过95%的IGF-I和IGF-II结合到6种特异性的IGF结合蛋白(IGFBP-1到6)。IGFBP不仅通过阻止IGF结合到细胞表面的受体以调节IGF的活性,而且某些IGFBP拥有不依赖于IGF的独立作用。IGFBP中含量最丰富的是IGFBP-3,正常血清中有超过75%的IGF与IGFBP-3结合。IGFBP-3是一种含有264个氨基酸的糖蛋白(去糖基后分子量为 29kDA) ,在循环系统中主要以高分子量的三聚体(包含IGF和酸敏感亚单位)形式存在。IGFBP-3的循环浓度取决于生长激素(GH)的充足程度,随年龄、青春期和性别而变化,并受蛋白酶活性调节。孕期妇女的血清中,蛋白水解活性显示出较高水平。GH不足时IGFBP-3水平较低,反之则升高。血清IGFBP-3检测在GH相关疾病的临床评估及治疗监测中具有公认意义。
【样本要求】
使用血清样本。采集血样后应尽快分离,-20°C下可长期储存,避免样本反复冻融。
