艾狄斯®尿胶原降解产物α链测试试剂盒（酶联免疫法）(ALPHACrossLaps^® EIA)（简称：αCTX-I检测试剂盒）采用酶联免疫吸附测定法定量测定人尿液中I型胶原C端肽（αCTX-I）的非异构化片段。

作为非异构化骨胶原降解的指标，αCTX-I检测试剂盒可用作鉴定乳腺癌和前列腺癌患者的骨转移的辅助工具。

此外，αCTX-I检测试剂盒可应用于变形性骨炎的诊断及相应抗骨吸收治疗疗效监测的辅助工具。

背景知识

n 尿胶原降解产物α链作为非异构化骨胶原降解产物的指标剂，可用于鉴别早期乳腺癌和前列腺癌骨转移患者，也可用于辅助诊断变形性骨炎及监测抗骨吸收治疗疗法患者。

n尿胶原降解产物α链检测试剂盒基于高特异性的抗氨基酸序列EKAHDGGR的单克隆抗体制备。为获得特异信号，氨基酸序列EKAHDGGR的两链必须相互交联。

【检测的概述和说明】

I型胶原占骨有机基质的90%以上，并主要在骨骼中合成。骨骼更新时，I型胶原被降解，短肽片段进入到尿液。这些片段可采用αCTX-I检测试剂盒来测定。

据文献报道，本试剂盒有助于评估变形性骨炎（1）骨吸收及检测前列腺癌(2，5) 和乳腺癌(3，4，5，6)骨转移。 

【检验原理】

αCTX-I检测试剂盒基于高特异性的抗氨基酸序列EKAHDGGR的单克隆抗体制备。为获得特异信号，氨基酸序列EKAHDGGR的两链必须相互交联。将标准、对照或未知的尿液样本移入涂有链霉亲和素的微滴孔中，然后应用生物素化抗体和过氧化物酶偶联抗体的混合物。然后，在抗体交叉抗原、生物素化抗体和过氧化物酶偶联抗体之间生成复合物，该复合物通过生物素化抗体与链霉亲和素表面结合。在2 – 8℃进一步孵化后,洗板并去除多余的洗液。加入显色底物，用硫酸终止止显色反应。最后测量吸光度。
